Insulin Separation Stationary Phase Comparison

BÜCHI Labortechnik AG

Successful peptide purification depends on selecting the right stationary phase. This application note evaluates the separation of insulin from small-molecule formulation components using flash chromatography and preparative HPLC on the BUCHI Pure Excellence C-950. The study compares different stationary phases and demonstrates how pore size, surface area and stationary-phase chemistry affect chromatographic performance.

Results show that conventional C18 phases did not achieve baseline separation, while wide-pore stationary phases significantly improved the separation of insulin from formulation excipients. The findings highlight the importance of stationary-phase selection when purifying peptides and other biomolecules.

Inside the Application Note:

  • Comparison of flash chromatography and preparative HPLC for insulin separation.
  • Evaluation of C18 and C4 stationary phases with different pore sizes and particle sizes.
  • Impact of pore size, carbon content and surface area on peptide retention and resolution.
  • Separation of insulin from formulation components such as metacresol and phenol.
  • Guidance for selecting stationary phases for peptide and protein purification workflows.

This application note is valuable for scientists working with peptide purification, preparative chromatography, pharmaceutical development and chromatographic method optimization.
